It's clear that the development of the next Mass Effect title (a Frostbite-developed title most likely meant for the next generation of consoles) is in extremely early stages, as the team allegedly still hasn't decided for the timeline of the story, and brings the question to the fans through exec producer Casey Hudson's Twitter account:

Parsing through your thoughts on the next #ME game. Would you be more interested in a game that takes place before the trilogy, or after?

A prequel wouldn't run the risk of invalidating previous story choices, so if it was up to me I'd go for that, but obviously there are arguments for both sides.
