So you've conquered the mighty Imperial sewers, and have stepped out into the wide expanse (depending on how your graphics are setup) of Cyrodiil, anxious to deliver the amulet the Emperor gave you. As I mentioned at the end of the tutorial, you have options, so take a moment to consider them. First, did you save your game just inside, and keep it separate? It will prevent you from having to redo the entire tutorial when creating a new character.

Weynon Priory, where your goal is to find Jauffre, is off to the west, following the main road from the Imperial City. You can of course open your map, and since you've heard about the location, click on it and fast travel right there, however I recommend taking the first trip on your own for the experience of travelling across the world on foot. Right now you're on the northeast side of the island which holds the Imperial City. Head west and wrap around the island, defeating the mudcrabs and wildlife, until you come to the western gate of Imperial City. On your way, keep an eye out along the northern shore (and anywhere else) for a green three-leaf plant called Nirnroot. Pickup any you can find, for they'll be useful later (though not related to the Main Quest).

At the western end of Imperial City you can either enter and take a look, or follow the road west northwest up toward Weynon Priory. Along the way you'll pass underneath a Fort on the way (that you can explore if you choose) and you may come across a bandit on the other side of the tunnel that demands payment for access. I chose to dispatch him, though it was a tough battle. In cases like this with tough opponents, don't be afraid to run while your magic is replenishing and then turn around to get a few more quick strokes in or deal damage. You can also heal yourself on the run. If you're unsure about your combat prowess, you may consider payment.

Soon enough, as you climb the rather steep incline for a road, you'll come to Weynon Priory (and be notified of that on your screen). It's a very small religious settlement near the bigger town of Chorrol. All towns in the game are gated and require a loading screen, even though you can see "in" them well enough from outside at certain vantage points.

As I mentioned in the walkthrough introduction, NPC's in Oblivion all have schedules they follow, similar to our own. During the day, most will be at their "job" and during the night they'll go back to bed. Sometimes their job gives them other duties and they'll be in one of a couple different places. They will always realistically travel between places as well, so you'll see them actually walking to and from their activities. In the case of Jauffre, if it's after the morning during the daytime, you'll find him in the large house up the righthand staircase behind a desk. If it's before, check the Priory (the tall building) and speak with Prior Maborel to ask where he can be found.

When you find Jauffre, give him the amulet, and go through all of the topics of conversation, eventually gaining his trust. Congratulations, this kicks off the next segment, Find the Heir, begun by continuing to converse with Jauffre. After the conversation, he gives you access to the nearby chest, so pillage and wear anything you can handle, and take anything else that may be of value when you head to Chorrol (hint, hint) to sell and explore.
