Ok, if you are a Wizard:
Target DC = 10 + your INT bonus + Level of the spell + any feat modifiers.
So if you have Spell Focus, it adds 2 to the target DC.
Example: Fireball cast by a Wizard with Spell Focus (Evocation) and Int 16. The DC = 10 + 3 + 3 + 2 = 18.
If you are a Cleric, Druid, Ranger or Paladin, substitute WIS for INT. If you are a Bard or Sorcerer, substitute CHA for INT.
There are three types of saves: Fortitude (CON based), Reflex (DEX based) and Will (WIS based). These are the numbers added to the targets Save roll.
So if a Fighter with a Reflex save of + 4 was trying to avoid the Fireball cast above, they would have to roll a 14 (on a d20) to beat the DC of 18.
Fortitude negates means that if the target makes their save, they are not affected in any way by the spell. Fortitude partial means that they take half damage from the spell. This is the same for Reflex and Will saves.
