I think it may just be in an already opened box. I've recently done that quest with a Mage and I completed the quest. I don't recall exactly where it was located though, but it doesn't require the main character to be a specific class, that's for sure.

If you move your cursor to the far right of the bar you can click and drag it along to make it bigger.

As for debuffs on enemies, I believe most debuffs have a graphical effect, so keep an eye out for that as to the duration.
