You can't make up your own quests in Fable. In fact, no RPG I can think of allows you to do this.

There's few, for example : The elderscrolls III : Morrowind, And TES IV: Oblivion.
Its because the Elderscrolls construction set.

But I think its not possible in fable.

Ahem. You forgot NWN2 and its fantastic toolset.

But as for Fable: TLC I don't think you can create any new questlines.

But that is because you need to have some modding tool to do that, right? If it doesn't exist, can you make up your own quest?

It doesn't exist in Fable. The only people with the power to craft their own quests are probably the game devs themselves.
