No one here will really endorse that last line of questioning. Just so you know.

Well I know a bit about virtual drives. There may be something going on with the disc drives. Where is your current drive at? Some games insist that you play the game with the disc in the same drive you installed it from.

Note that emulation software can cause the order of drives to be changed a bit. For example, you install the game from the D: drive. In using emulation software later on, D: could have been changed into E:. If the virtual drive D: is removed at a later point, the real CD drive could still be at E:. The game might mistakenly think it's being started from the wrong drive.

That's the only other thing I can think of. My best guess is still a faulty registry entry though. DX2 is especially notorious for taking bad care of it's own registry, as the various community texture packs can cause all kinds of problems if they're not uninstalled correctly before you uninstall DX2.

You seem to have removed them manually. Perhaps a registry cleaner tool can be a bit more thorough.

Edit: Did you try reinstalling the game >after taking care of the registry entries?
