So here I was beginning to think I couldn't detect it because I am playing a Paladin and went through the side mini-quest to get the Avenger sword, and thought maybe the treasure room secret door was keyed so that you can't do both.
What's frustrating is my character can disable traps in that room with the sceptre, and cast sound burst into the room (yes, past the closed / non-existent door) using the horn of blasting, but still can't detect the door itself!
